Nace en Belfast en 1970, y compagina su formación multidisciplinar con el interés político por la problemática de su tierra, Irlanda del Norte. A los 19 años se sale por primera vez del buen camino: debuta como guionista con historietas cortas para la revista británica Crisis, y da el salto a publicaciones como 2000 AD y Judge Dredd Magazine. A raíz de estos trabajos conoce a artistas británicos como John McCrea y Steve Dillon, sus futuros cómplices. En 1991 entra en el mercado norteamericano con guiones para la serie más veterana de Vertigo: Hellblazer. En 1996 y con la ayuda de Steve Dillon, crea a Jessie Custer y nos muestra su particular visión del mundo en Predicador. Otras obras de Ennis son Hitman, Dicks, El Soldado Desconocido o War Story. Desde 2002, trabaja para WildStorm, y en 2006 inicia la publicación de The Boys, una de sus series más mordaces, explícitas y exitosas.

Su vocación historietística fue temprana y mientras estudiaba dibujo y diseño en Belfast movía incansablemente su trabajo por convenciones de cómic y editoriales inglesas, como 2000 AD o Marvel UK. Así, pronto comenzó a realizar algunos pequeños trabajos profesionales en algunas revistas y llegó a colaborar finalmente en 2000 AD, dibujando algunas historias de Future Shocks. Sus trabajos tempranos están notablemente influenciados por John Byrne y Alan Davis.
